Determination of seven kinds of flavone aglycones in red wine with HPLC-UV and their bioactivities

Abstract

Quantitative determination of quercetin,morin hydrate,kaempferol,isorhamnetin,luteolin,apigenin in red wine with high performance liquid chromatography(HPLC) with ternary gradient was established.The flavonoids were separated by HPLC with Luna C18(4.6mm×150mm,3μm)and a Phenomenex C18 precolumn(4mm×3.0mm) at 50℃.Mobile phase A,B and C were acetonitrile,ethanol and 0.2% H3PO4,respectively.Gradient elution rate was 0.25ml/min and detection wavelength was 265nm.The results showed that the calibration curves were linear at ranges of 0.7mg/L~32.0mg/L.The average recovery in the between of 98.2%~101.1% and RSD 1.88%~2.85%.The method is simple and accurate with good reproducibility and can be used in quantity determination of flavonoid aglycone of natural product.
