Abstract
Objective: To investigate the protective effects of paeoniflorin(PF) on PC12 cells injury by 6-hydroxydopamine(6-OHDA) and its possible mechanism.Methods: PC12 cells were treated with 6-OHDA to set up the damaged cell model.Cell survival was detected by MTT and lactate dehydrogenase.The mRNA expression of Bcl-2 and Bax were detected by RT-PCR.The apoptosis was monitored with Hoechst 33342/PI staining.Results: Paeoniflorin can significantly decrease 6-OHDA-induced LDH leakage rate and apoptosis.Paeoniflorin could increase the expression of Bcl-2 mRNA,and decrease the expression of Bax mRNA.Conclusion: Paeoniflorin inhibited 6-OHDA-induced apoptosis in PC12 cells.The mechanism is related to the regulation of mRNA expression of Bcl-2 and Bax in PC12 cells.
