Abstract
OBJECTIVE To investigate the effect of glutamate (Glu) on cultured PC12 cells and the protective effects of paeoniflorin.METHODS Cell survival was dotained by MTT and lactate dehydrogenase,cell apoptosis was measured by flow cytometry,the decreasing concentration of [Ca~ 2+ ]_ i was dotained by Fura3/AM and the mitochondral membrance potential in PC12 cells was dotained by flow cytometric assay.RESULTS PC12 cell damage, the concentrations of [Ca~ 2+ ]_ i , and apoptosis induced by Glu were decreased after being administrated with paeoniflorin. CONCLUSION Paeoniflorin inhibits Glu-induced apoptosis in PC12 cells. The mechanism is related to inhibiting intracellular Ca~ 2+ overload and maintaining mitochondral membrance potential.
