Abstract
Objective To study the expression of Oct4,Nanog protein in human non small cell lung carcinoma and normal lung tissue by tissue chip technique and to analyze their clinical significance. Methods The tissue microarray technique was applied to prepare samples of 186 cases of lung carcinoma and 23 cases of normal lung tissue. The expressions of Oct4 and Nanog protein were detected by using immuno-histochemical assay. Results The positive expression rates of Oct4 and Nanog were 46.8%(87/186) and 81.7%(152/186) respectively in non-small cell lung cancer tissue, while they were 0(0/23) and 30.4%(7/23) respectively in normal tissue, and the expressions of Oct4 and Nanog protein in carcinoma tissue were significantly higher than those of normal tissue(both P0.01).The expressions of Oct4 and Nanog protein were significantly correlated with lymph node metastasis and clinical stages, but they were not correlated with gender, age, tumor size and histological classification. Conclusion Oct4 and Nanog play a key role in pathogenesis of NSCLC.Combined detection of Oct4 and Nanog may serve as important parameters to evaluate the metastasis and malignant grade of the malignancy.
