Abstract
Objective To investigate the effects of angiotensinⅡ(AngⅡ) and angiotensinⅡ type 1 receptor antagonist (AT1RA) on the collagen Ⅲ synthesis and collagen Ⅲ mRNA expression of in vitro cultured hepatic stellate cells (HSCs). Methods HSC-T6 rat hepatic stellate cell line was chosen as the study model of the activated hepatic stellate cells. Cultured HSCs were randomized into control group, AngⅡgroup, AT1RA group and AngⅡ+AT1RA group. Cell culture supernatant was used to detect collagen Ⅲ level by ELISA. HSCs were harvested to measure the collagen Ⅲ mRNA expression by RT-PCR. Results Collagen Ⅲ level of control group, AngⅡgroup and AngⅡ+AT1RA group in cell culture supernatant was(6.301±0.432)ng/ml,(7.356±0.237)ng/ml and(6.263±0.236)ng/ml respectively, AngⅡgroup was higher than control group(P0.05), AngⅡ+AT1RA group was lower than AngⅡgroup(P0.05). Collagen Ⅲ mRNA expression level of control group, AngⅡgroup and AngⅡ+AT1RA group in HSCs was 2.317±0.015, 2.643±0.057 and 2.330±0.036 respectively, AngⅡgroup higher than was control group(P0.05), AngⅡ+AT1RA group was lower than AngⅡgroup(P0.05). Conclusions AngiotensinⅡ can increase the collagen Ⅲ synthesis and collagen Ⅲ mRNA expression of hepatic stellate cells, while all these effects are inhibited by angiotensinⅡ type 1 receptor antagonist.
