Abstract

Objective To investigate the safety and therapeutic efficacy of intratumoral injection of p53 gene (Gendince~) or bronchial artery infusion with p53 gene combined with bronchial artery infusion (BAI) in patients with lung cancer. Methods Fifteen patients suffering lung cancer confirmed by pathology were percutaneously injected with p53 gene into the tumor lesions under the guidance of CT or infused with p53 gene into bronchial artery via a catheter. Two to five days after the procedure, the patient was treated with BAI and the chemical drugs were cisplantin, 5-Fluorouracil and/or etoposide. Then, p53 gene therapy or BAI was carried out repeatedly according to the condition of the patient. Therapeutic efficacy was evaluated with CT scan. Results The combined therapies were accomplished in all patients and the follow-up time ranged 2-11 months. After p53 gene therapy, 6 patients suffered fever ranging 38-40℃ with few and mild other side effects of it observed. In the course of the combined therapy, no severe complications appeared. Of all 15 patients, the total effect rate (CR+PR) was 46.7%(7/15) with tumor lesion disappearance in 1 case, significant tumor regression in 6 cases, mediastinal lymph nodes shrinkage in 3 cases, regression of thoracic fluid in 1 case and 93.3% (14/15) of the cases had alleviation of clinical symptoms. Conclusion The study preliminarily revealed that p53 gene combined with BAI, as one of the comprehensive therapies for tumor treatment, is safe and effective in lung cancer.
