Mechanism of matrine suppressing HT-29 cell line proliferation

Abstract

Objective To study the anti-tumor mechanism of matrine by observing the effect of matrine on HT-29 cell line proliferation and apoptosis.Methods Using cells culture,MTT,FCM,TRAP methods,the effects of matrine on the proliferation,apoptosis and telomerase activity of HT-29 cells were analyzed.Results After treatment for 24 h by matrine(0,3,6,8,10 μg/L),the inhibitory rate of proliferation was 0,(33.7%,)(59.0%,)(66.7%,)69% respectively.After the HT-29 cells were cultured with 5 μg/L matrine for 0,24,48,72 h,the apoptosis rate was(0.85%,)(15.31%,)(17.99%,)(26.58%,)respectively.The telomerase activity was decreased with prolongation of matrine treatment time.Conclusion Matrine has the anti-tumor effect by suppressing HT-29 cells proliferation through inhibiting DNA synthesis,inducing apoptosis,inhibiting telomerase activity.
