Cadmium induced apoptosis in rat hepatocytes and cytoprotective effect of N-acetyl-cysteine.

Abstract

Rat hepatocytes were isolated by a two-step perfusion technique.Applying MTT assay,Hoechst 33258 and flow cytometry detected hepatocytes cell viability and apoptosis rate induced by cadmium acetate.And the effects of N-acetyl-cysteine(NAC) on cell viability and apoptosis rate were investigated.The results showed that cell viability were decreased(P0.01)and apoptosis rate were increased(P0.05)in a dose-dependent manner after exposure to 2.5,5 and 10 μmol/L cadmium acetate.2 mmol/L NAC increased cell viability(P0.01)and decreased apoptosis rate.It suggests that cadmium acetate may induce the apoptosis of hepatocytes at certain doses and NAC has cytoprotective effect on cadmium.
