Tissue culture of the different explants of Lycoris aurea Herb.

Using wild medicinal plant Lycoris aurea Herb.as the experimental materials,eight kinds of explants from L.aurea Herb.were cultured on two kinds of culture medium:MS+10 mg·L-1 BA+5 mg·L-1 NAA+0.7% agar+3% cane sugar,and MS+5 mg·L-1 BA+10 mg·L-1 NAA+0.7% agar+3% cane sugar.The effects on tissue culture of the eight kinds of explants were studied.The results showed that the adventitious bud rate,adventitious root rate,adventitious bud and root rate,and browning rate of the double endothecium's bulb-scale explants cultured on the medium of the concentration for 10 mg·L-1 BA+5 mg·L-1 NAA were 90.33%,87.00%,85.00% and 6.33%,respectively.Those rates of the same explants cultured on the medium of the concentration for 5 mg·L-1 BA+10 mg·L-1 NAA were 46.67%,82.33%,35.33% and 16.67%,respectively.Those rates above were significantly different with those of other explants;The correlation analysis showed that the effect of the eight kinds of explants on one kind of the medium was significantly correlated with that of itsself on another kind of the medium.Both Duncan's multiple range test and correlation analysis indicated that the effect of the double endothecium's bulb-scale explants cultured on the medium of MS+10 mg·L-1 BA+5 mg·L-1 NAA+0.7% agar+3% cane sugar was the best and the double middle-layer bulb-scale explants cultured on the same medium was next.
