PTEN mRNA and PTEN protein expression and their significance in endometrial adenocarcinoma.

Abstract
[Objective] To study the expression of PTEN mRNA and PTEN protein in endometrial adenocarcinoma.[Methods] The expression of PTEN mRNA and protein were examined by RT-PCR and immunohistochemistry for 40 cases with endometrial adenocarcinoma,18 cases with endometrial atypical,18 cases with endometrial hyperplasia and 18 cases normal endometrial,respectively.Cell apoptosis in each endometrial tissures was determined by flow cytometry.[Results](1) PTEN expression level of mRNA in patients with endometrial carcinoma and endometrial atypical hyperplasia was significantly lower than those patients with endometrial hyperplasia and normal endometria(P﹤0.05).The deletion of PTEN mRNA expression in endometrial adenocarcinoma,endometrial atypical hyperplasia,endometrial hyperplasia and normal endometrial tissures were(29.8 ± 4.6) %,(43.2 ± 3.5) %,(1.7 ± 1.0) % and 0%,respectively.(2) The PTEN protein expression deletion in endometrial adenocarcinoma and normal endometrial tissures were 60.0% and 0%,respectively,there was significance difference between them(P ﹤ 0.01).The PTEN protein expression deletion in endometrial carcinoma was correlated to clinical stage and histological grading(P﹤0.05) and compared between endometrial hyperplasia and normal endometrial tissues and endometrial carcinoma,endometrial atypical tissues there was significant difference of apoptosis rate(P﹤0.05).[Conclusion] The PTEN mRNA and PTEN protein expression deletion may play an important role in the genesis and development of endometrial carcinoma and may be as a diagnostic biologic mark of early endometrial adenocarcinoma.
