Abstract
The extra-cellulase from T.reesei QM9414 was immobilized on chitosan by glutaraldehyde,then the activity,optimum pH,Km,and optimum temperature of immobilized and free cellulose was also studied.The results indicated that protein concentration and enzyme activity of free enzyme was 33.3 ug/mL and 120IU/mL,respectively.The optimum pH of soluble enzyme and immobilized enzyme were pH 5 and pH 4,respectively.The optimum temperature of soluble enzyme was 50℃,whereas immobilized showed high activity in 60℃.The Km of immobilized enzyme and soluble enzyme was 3.592 01×10-2 and 4.076 04×10-2,respectively.
