Abstract

Objective: To study the growth inhibitory effect of trichostatin A (TSA) on non-small cell lung cancer A549 cells and its mechanisms. Methods: A549 cells were exposed to different concentrations of TSA(0.1μM, 0.5μM and 1μM)for 24h. MTT assay was performed to measure the inhibition of cell proliferation. Cell cycle distribution was analyzed by PI staining. P21 protein expression was detected by western blotting. The mitochondrial membrane potential (MMP) was measured by flow cytometry. Apoptosis was analyzed with Annexin V-PI staining by flow cytometry. Results: The results showed that TSA inhibited cell growth in a dose dependent manner. TSA induced G2/M arrest, which was accompanied by increased level of P21 protein expression. TSA caused increasing loss of MMP and apoptosis dose-dependently. Conclusions: These results suggest that TSA inhibits cell growth by causing G2/M cell cycle arrest and apoptosis.
