Simultaneous determination of four effective compounds in Juejin Decoction by high performance liquid chromatography

Abstract

A simple method using high performance liquid chromatography coupled with ultraviolet (UV) detection was developed for the separation and determination of 5-Hydroxymethyl furfural, Ferulic acid, Cinnamaldehyde and Cinnamate acid in Juejin Decoction. The chromatography was performed on a C18 column at 30 °C, with a mobile phase composed of water containing 0.02% glacial acetic acid and methanol with gradient elution. The flow rate was 1.0 ml/min and UV detection wavelength was 282 nm. The linear ranges of the four compounds were 2.70-27.0 μg/ml (r=0.9996), 49.7-497 μg/ml (r=0.9995), 5.55-55.5 μg/ml (r=0.9996) and 0.359-3.59 μg/ml (r=0.9999), respectively. The recoveries of the four compounds were in the range of 98.2%-99.0%. The method is rapid, accurate and suitable for simultaneous determination of four effective compounds in Juejin Decoction.
