Abstract

Rats pretreated with daily injections of insulin display, during the period immediately following cessation of injections, symptoms indicative of insulin insufficiency. During this interval, hepatic glucose-6-phosphatase levels are consistently elevated. Administration of tolbutamide (250 mg/kg body weight per os) fails to cause a depression in hepatic glucose-6-phosphatase levels in these animals, whereas, in normal rats, this agent does reduce significantly the levels of activity of this enzyme in the liver.The failure of tolbutamide to depress hepatic levels of glucose-6-phosphatase in insulin-pretreated rats in which secretion of insulin cannot be readily obtained is consistent with the hypothesis that one of the means by which tolbutamide exerts its effects is stimulation of the release of this hormone by the islets of Langerhans.
