Correlation between the TLR4 mRNA levels in peripheral blood mononuclear cells and HBV DNA in hepatitis B-virus infected subject.

Abstract
Objective To investigate the correlation between the TLR4 mRNA levels in peripheral blood mononuclear cells(PBMC) and hepatitis B virus DNA from HBV infected patients.Method TLR4 mRNA levels in PBMC was measured by RT-PCR,and the HBV DNA viral load of hepatitis B virus infected patients was measured by using real time fluorescence quantitative PCR.The correlation between the TLR4 mRNA levels and HBV DNA of the patients was analyzed.Result TLR4 mRNA level in three groups with different viral load(1×103copies/μg DNA;1×103copies/μg DNAand1×105copies/μg DNA;1×105copies/μg DNA) were significantly different(P0.01).The log values of HBV load in all patients had negative correlation with the TLR4 mRNA levels in their PBMCs (r=-0.537,P0.01).Meanwhile the TLR4 mRNA levels had positive correlation with ALT and AST(r=0.608,r=0.659,P0.01).Conclusion The active replication of HBV DNA and higher level of HBV DNA copies in hepatitis B virus infected suggested that they have relation with the lower expression of TLR4 mRNA in PBMC.
