Abstract
Thrombopoietin (TPO), the endogenous Mpl ligand, maintains peripheral platelet concentrations at a constant level by modulating megakaryocytopoiesis to compensate for changing peripheral requirements (1–6) . TPO promotes platelet production by inducing hemopoietic stem cells to differentiate into megakaryocytes (3,7–9) , and stimulating megakaryocyte proliferation and endoreduplication (3,10–16) , thereby amplifying formation of the megakaryocyte cytoplasmic substrate destined for fragmentation and release as functional circulating platelets (17–19) (see Chapters 14–17).
