Cloning and sequencing analysis of the 5′ fragment of GPAT gene from Citrus grandis cv. Xingantezaoyou.

Abstract

Citrus maxima cv.Xingantezaoyou's GPAT gene of 5' end fragment was isolated from the leaf by means of 5'-RACE technology.The cDNA fragment is 832 bp long(GenBank Accession No:DQ193970).The nucleotide sequence analysis showed that these sequences of GPAT 5' end fragment are greatly different from those of other plants.The best nucleotide similarity to Citrus unshiu was 98.9% but those Carthamus tinctorius,Arabidopsis thaliana,Elaeis guineensis,Pisum sativum and Vicia faba were 62.5%,54.6%,48.2%,47.4% and 46.9% respectively.The deduce amino acids sequence analysis showed the similarity to Carthamus tinctorius,Arabidopsis thaliana,Elaeis guineensis,Pisum sativum and Capsicum annuum were 51.6%,60%,45.1%,48% and 52.3%.The amino acid sequences of GPAT gene has genera specificity because the similarity of amino acid sequences to Bacillus amyloliquefaciens is only 18.9%.
