Abstract

Abstract The effective transdermal drug delivery depends on the capability of the formulation to deliver the drug through the skin at the amount adequate to exert desired pharmacological reponse. However, the skin, especially stratum corneum is the main barrier for the permeation of substance.  The structure of the lipid in the stratum corneum is well organized, thus making the pentration through the stratum corneum a rate limiting step for most substraces. Variousresearches attempted to find the method to enhance skin permeation.  Skin permeation enhancer is one of the popular methods used in transdermal formulations.  Among various substances evaluated for skin penetration enhancers, fatty acid is one of the promising substances.  In this review, fatty acid as a skin permeation enhancer is elaborated including its skin enhancing property, mechanisms of action, and factors affecting the skin permeation.
