An epidermal growth factor receptor (EGFR) tyrosine kinase inhibitor (TKI) is the preferred first-line treatment of choice for patients with *EGFR*-mutated advanced non-small cell lung cancer (NSCLC), with gefitinib, erlotinib, afatinib, dacomitinib and osimertinib being commonly used in clinical practice. About 10–20% of patients with NSCLC patients harbor uncommon *EGFR* mutations with varying sensitivity to different EGFR-TKIs. This review presents an overview of the latest clinical data on the potential treatment strategies for NSCLC harboring uncommon *EGFR* mutations.
