Abstract
Objective To study the effect of olmesartan on the expression of AT1 receptor mRNA in rostral ventrolater medulla(RVLM)of the spontaneously hypertensive rat (SHR). Methods Rats were divided into SHR contral group (normal drinking),SHR-Olme group [(olmesartan,30 mg(/kg·d)],and Wistar-Kyoto rats(WKY)group (normal drinking). Four weeks later,these rats were decapitated to death and got the brain tissue. AT1 receptor mRNA level in RVLM was measured by RT-PCR and analyzed with optic density. Results The blood pressure of all SHR-olme rats reached normal level after administration of olmesartan for a week. The mean optic density(MOD) of AT1 receptor mRNA in RVLM of SHR-olme was much reduced compared with SHR group[(0.94±0.41)vs(2.41±0.37),P 0.01],but higher than that of WKY group(0.81±0.22,P 0.05). Conclusion Olmesartan could singificantly reduce the leve of AT1 receptor mRNA in RVLM of the SHR.
