Abstract

Objective:To investigate the effect of arsenic trioxide (As 2O 3) on proliferation, cell cycle regulation and apoptosis in leukemic cell lines and it's mechanism. Methods:MTT assay, electrophoresis of genomic DNA,as well as protein/DNA multiparameter flow cytometry were used. Results:Cell proliferation were significantly inhibited in a dose and time dependent manner.Apoptosis were induced in these cells.Furthermore,As 2O 3 could change the content of some cell cycle and apoptosis related proteins in NB 4/Jurkat cells in which both G 1 and G 2/M phases were arrested. Conclusion:The change of all cycle regulatory protein may play an improtant role in proliferation inhibition and apoptosis of leukemic cells regulatery from As 2O 3.
