Effect of pyrrolidinecarbodithioic on rats with severe acute pancreatitis

Abstract

Objective To investigate the effect of pyrro-2-lidinecarbodithioic (PDTC) on rat with cute necrotizing pancreatitis (ANP).Methods Thirty-six Wistar rats were randomly divided into 4 groups:sham operation group (n =6),sham operation plus intravenous injection group (n =6),ANP group (n=12)and ANP plus intravenous injection group (n=12).ANP were induced by retrograde injection of 3% sodium taurocholate (0.1 ml/100g weight) into the biliopanereatic duct.Rats of sham operation group only underwent opening the abdomen and rubbing the pancreas.Rats of intravenous injection group received intravenously 25 mg/kg body weight PDTC.After 6h of establishment of the ANP model,the rats were sacrificed and the serum levels of amylase and lipase were determined.Pancreatic tissues were examined for pathological score.The activation levels of NF-κB and apoptosis index of pancreatic cells were detected by immunohistochemistry.Results The serum levels of amylase in sham operation group,sham operation plus intravenous injection group,ANP group and ANP plus intravenous injection group were (3121±34)U/L,(2985±89)U/L,(7422±99) U/L and (5330±94) U/L,respectively;while the serum levels of lipase were (33±11) U/L,(36±11)U/L,(566±167) U/L and (363±132) U/L,respectively;pathological scores were 0,0,22.13±0.61 and 17.38±0.53,respectively;the positive rates of NF-κB were 0,0,(66.7±4.2) % and (38.6±2.7) %,respectively;apoptesis index were 0,0,(7.80±0.56)% and (13.3±0.73)%,respectively;all the values in the ANP group were significantly higher than those in sham operation group (P<0.05),PDTC injection alone had no effect on all the values,however,after PDTC injection,all the values were significantly lower than those in ANP group (P<0.05).Conclusions PDTC could alleviate pancreatic tissue damage possibly by inhibiting the activation of NF-κB,accelerating the apoptosis of pancreatic ceils and reducing release of pancreatic enzyme.
