Generation and characterization of liner B-cell epitope monoclonal antibody in the hemagglutinin of human-infected avian-origin H7N9 influenza virus

Abstract

Objective To obtain and characterize the monoclonal antibody (McAb) of liner B-cell epitope in H7N9 influenza virus hemagglutinin. Methods Six weeks old BALB/c mice were immunized with synthetic peptide of linear B-cell epitope of H7N9_H7 and McAbs were prepared by hybridoma technology. The synthetic peptide ELISA method and Western Blot method were established to detect McAb subtype, specificity and purity. Results Two hybridoma cell lines, 7C8H8 and 8E9E2, stably excreting anti-H7N9_H7 McAb were obtained. 7C8H8 was IgG2a, and it had higher titer (> 1: 512 000) and specificity. 8E9E2 had the weaker binding ability with H7N9_H7. Conclusions By comprehensive various techniques, one anti-H7N9_H7 McAb7C8H8 is obtained, and have higher titer and specificity.
