Abstract
Objective To investigate the effects of hyperbaric oxygen (HBO) preconditioning on inflammatory reaction induced by ischemia-reperfusion (IR) in rats following skin flap transplantation.Methods Fifty-six Sprague-Dawley rats were randomly divided into 7 groups:the sham group,the ischemia-reperfusion (IR) day 1 group,the IR day 3 group,the IR day 5 group,the HBO preconditioning + ischemia - reperfusion day 1 group,the HBO + ischemia-reperfusion day 3 group,and the HBO + ischemia-reperfusion day 5 group.The abdominal pedicle skin flap animal model was established and the animals received HBO preconditioning.The levels of neutrophil elastase(NE) and high mobility group box 1 ( HMGBI ) of 7 animal groups were measured with ELLSA.Results The levels of NE in the IR day 1,day 3 and day 5 groups were obviously higher than those of the sham group.The NE level of the HBO preconditioning + IR day 1 group was obviously lower than that of the IR group ( P < 0.01 ),and the NE levels of the HBO preconditioning + IR day 3 and day 5 groups were lower than those of the IR group(P <0.05).The levels of HMGB1 in the IR day 1,day 3 and day 5 groups were significantly higher than that of the sham group (P<0.01).The HMGB1 levels of the HBO preconditioning + IR day 1 and day 3 groups were lower than those of the IR group ( P < 0.05 ).The HMGB1level of the HBO preconditioning + IR day 5 group was obviously lower than that of the IR group ( P < 0.01 ).Conclusions Experiments showed that HBO preconditioning could decrease the levels of NE and HMGB1 in rats following skin flap transplantation,alleviate inflammatory reaction induced by ischemia-reperfusion following skin flap transplantation,and increase survival rate of the grafted skin flap.
