Abstract
Objective To evaluate the expressions of Caspase-3, Caspase-8, Caspase-9 and Bax, Bcl-2 under the intervention of HPS-1 in A549 cells. Methods After treated by HPS-1, the viability of cell lines was detected through methylthiazolyldiphenyl-tetrazolium bromide (MTT). The apoptosis of cell lines was detected by flow cytometry (FCM). The expressions of Caspase-3, Caspase-8, Caspase-9 were measured by colorimetry. Results The results showed that different doses of HPS-1 inhibited the proliferation and promoted the apoptosis of A549 cells.And the expression levels of Caspase-3, Caspase-8, Caspase-9 proteins increased in a concentration-dependent manner in A549 cells treated with HPS-1, and these effects were significantly enhanced in A549 cells treated with high does of HPS-1. Conclusions HPS-1 has a powerful effect on inhibiting cellular proliferation and inducing cellular apoptosis of lung adenocarcinoma A549 cells, which may be associated with the increased protein proportion of Caspase-3, Caspase-8, Caspase-9 and expression of Bax mRNA, decreased expression of Bcl-2 mRNA.
