Abstract
Objective To study the effects of Ursolic Acid (UA) on the Human MC63 osteosareoma cell. Methods Human MC63 osteosareoma cells were cultured in vitro,and the concentrations 10,20,50,100 μmol/L of ursolic acid were added into culture fluid. Cell proliferation were measured by methyl thiazol tetrazolium (MTT), the expression of mRNA levels of bone morphogenetic protein (BMP)-2 and BMP-6 were detected by reverse transcription-polymerase chain reaction (RT-PCR). And the expression of proteins were detected by Western boltting. Results Compared with the blank group, the proliferation in experiment group was suppressed with time and concentration dependent. The proliferation was 0. 25 ±0. 03in 100 μmol/L group while it was 0. 86 ± 0. 10 in 10 μmol/L group after 72 h ( P < 0. 05 ). The proliferation was statistical significance after 24,48,72 h in different concentrations. The expressions of BMP-2 and BMP-6 were higher than that in blank group, and there was statistical significance ( P < 0. 05 ). But there was no statistical significance between two experiment groups (P > 0. 05 ). Conclusion Ursolic acid can suppressed the proliferation of MG63 osteosareoma cells with time and concentration dependent. promoted,which has relation with promoting the expressions of BMP-2 and BMP-6.
