Abstract
Objective To observe the inhibitory effects of an antisense Survivin vector on proliferation and invasion of osteosarcoma cells line MG-63, and explore the mechanism. Methods MTT assay was used to measure the proliferation inhibition after transfection of MG-63 cells with Survivin antisense RNA. Boyden chamber in vitro invasion assay was applied to observe the changes of invasion capability.The expression of Angd-2 before and after transfection was detected by reverse transcription-polymerase chain reaction (RT-PCR). Results The proliferation of cells transfected with antisense Survivin was significantly inhibited ( the inhibition rate was 23.4% ) as compared with other groups. The invasion ability was remarkably decreased after antisense transfection ( 17. 9% ) as compared with the control groups. Moreover,the mRNA expression of Ang-2 in antisense Survivin transfected cells was much lower than control groups.Conclusion The antisense Survivin vector could markedly inhibit the proliferation and invasion of osteosarcoma cells line MG-63, and partially reversed their malignant phenotype, which was possibly related with down-regulation of the Ang-2 expression.
