Effect of RNA Interference on the Proliferation and Apoptosis of Acute Monocytic Leukemia Cells

Abstract

Leukemia is a malignant clonal disease of hematopoietic stem cells. Clonal leukemia cells proliferate andaccumulate in bone marrow and other hematopoietic tissues due to uncontrolled proliferation, differentiationdisorders, and blocked apoptosis, and infiltrate other non-hematopoietic tissues and organs, while inhibitingnormal hematopoietic function. The effect of RNA interference with MAGE-A3 gene on the proliferation ofleukemia cells was studied. Methods: the expression of MAGE-A3 gene and the proliferation of leukemia cellswere observed. the siRNA sequence that can effectively silence the expression of CDX2 gene in K562 wassuccessfully constructed and screened in this study. transfecting the siRNA into K562 cells can significantlyreduce cell proliferation and significantly promote cell apoptosis, suggesting that high expression of CDX2 isone of the reasons that lead to uncontrolled self-renewal and blocked apoptosis of leukemia cells. This study hasa certain reference value for the clinical treatment of leukemia.
