Abstract

Abstarct The study, to compare the modified trichrome staining methods and Wheatley’s trichrome staining methods which used of determine parasitic characteristics. A total of 77samples, which were determined as 9 negative and 68 positive by saline-iodine, modified asic fast and modified trcihrome staining, were stained with Wheatley’s trichromestaining method and examined by light microscopy with immersion oil. Positivity wasdetected at 25 of 77 samples stained by Wheatley’s trcihrome staining method. 43 samples which were detected as positive by staining modified trichrome staining method werefound to be negative by Wheatley’s trichrome staining method. Also 9 samples whichwere determined as negative by modified trichrome staining method were determinedas negative by Wheatley’s trichrome staining method. This study showed that modified trichrome staining method is more effective than Wheatley’s trichrome method stai-ning for microscopic diagnosis of amoebic trophozoites.
