Abstract
Objective:To investigate that the effect of adipose-derived stem cells(ADSCs)on immune function of C57 mice with thymomas induced by irradiation, and plausible mechanism. Methods:A total of 32 cleaning degree C57BL/6mice were divided into four groups:control group of 8mice with non-irradiation;irradiation group of 8mice with irradiation;irradiation+ADSCs group of 8mice,thymoma model mice were injected 0.5mL ADSCs via tail vein at one day after last irradiation;non-irradiation+ADSCs group of 8 mice with the same ADSCs injection as irradiation+ADSCs group.Mice in four groups were sacrificed at 7th day after last irradiation, thymus index was measured by weight and thymus lymphocyte transformation rate was detected by MTY.Thymus lymphocyte cycle and T lymphocyte subsets were determined by flow cytometry. Results:Compared with irradiation group mice, lymphocyte transformation rate and thymus index was significantly higher in irradiation+ADSCs group mice. When detected by flow cytometry,CD4+T cell subsets and CD4+/CD8+from irradiation+ADSCs group mice increased,CD8+T cell subsets from irradiation+ADSCs group mice was decreased, there were statistical significance(P0.05).Conclusion:ADSCs may regulate the cell levels of mice thymus and protect immune function of mice.
