EFFECT OF SODIUM FERULATE ON THE APOPTOSIS OF HUMAN UMBILICAL VEIN ENDOTHELIAL CELLS INDUCED BY OX-LDL AND EXPRESSION OF P53

Abstract

[Objective] To investigate the inhibition and mechanism of sodium ferulate(SF) on the apoptosis of human umbilical vein endothelial cells(HUVEC-CRL1730) induced by oxidized low density lipoprotein(ox-LDL).[Methods] HUVECs were induced injury by ox-LDL in vitro,and then treated with sodium ferulate.Cell viability was determined by MTT assay,morphology and apoptosis were observed by Hoechst33258 fluorescence staining,and cell apoptosis rate was detected by flow cytometry(FCM) assay.The expressions of p53 protein was determined by Western-blot.[Results] Ox-LDL could significantly inhibit HUVEC cells growth and proliferation;induce HUVEC apoptosis and markedly upregulate p53 protein expression level.But after treated with sodium ferulate,the cell cytoactivity increased obviously compared with the cells treated with oxLDL(P﹤0.05),the apoptosis rate decreased markedly(P﹤0.05) and p53 protein expression level downregulated significantly(P﹤0.05).[Conclusion] ox-LDL induce HUVEC cells apoptosis probably through upregulating p53 protein expression.But sodium ferulate could inhibit the apoptosis of HUVEC through its downregulating effect on the expression of p53 protein.
