Abstract

The effects of enzyme system,reaction time and temperature,osmotic stabilizer on protoplast formation and regeneration from Asp.niger were studied by orthogonal test.The optimal condition for protoplast formation and regeneration of Asp.niger was obtained.The concentration of protoplast formation was up to 30×106/mL and the protoplast regeneration rate was over 25% for the strain.The procedure of formation of protoplast for Asp.niger was observed under Olympus microscope.
