Abstract
Widal test is agglutination test for serological diagnosis of typhoid fever or enteric fever. Widal test measures the agglutination level of antibody towards O antigen (somatic) and H antigen (flagellar). The test is usually performed on acute serum, that is first serum which taken at the first time contact with patients, at least must be obtained 1 ml of blood to get enough serum. Widal testing material is serum, but due to the limited blood volume, while saving the examination time, many laboratories using plasma samples as an alternative. The research goal is to know the difference of Salmonella typhi-O and typhi-H titer using plasma and serum. The research type is analytical with cross sectional approach. The research towards 16 research samples were examined using serum and plasma materials. The research result was serum samples obtained negative Salmonella typhi-O, titer 1/80, titer 1/160, titer 1/320 consecutive as much as 2,6,6,2 samples. Salmonella typhi-H titer 1/80, titer 1/160, titer 1/320 consecutive as much as 1, 1,11,3 samples. Plasma samples were obtained. Negative Salmonella typhi-H, titer 1/80, titer 1/160, titer 1/320 consecutive as much as 1,7,6,2 samples. Salmonella typhi-H titer 1/80, titer 1/160, titer 1/320 consecutive as much as 1, 13, 2 samples. The statistic test result with Wilcoxon and was Chi Square obtained p > 0,05 which means there was no significant difference of Salmonella typhi-O and typhi-H serum and plasma samples.
