Hypoxia inducible factor-la in combination with bcl-xL siRNA suppresses growth of lung adenocarci-noma A549 cells

Abstract
Objective To explore the effect of hypoxia inducible factor 1 alpha (HIF-1 α) and bcl-xL small interfering RNA in combination on the proliferation of lung adenocarcinoma A549 cells.Methods A549 cells in vitro were divided into four groups:group A treated with normal saline,group B with HIF-1α siRNA,group C with bcl-xL siRNA,group D with HIF-1α siRNA and bcl-xL siRNA.The expression of HIF-1α and bcl-xL mRNA was detected by using reverse transcription-polymerase chain reaction (RT-PCR),their proteins were detected by using Western blotting,and MTT assay was used to detect the growth of A549 cells.Results In groups B and D,the expression level of HIF-1α mRNA was decreased to 0.733 ± 0.157 and 0.766 ± 0.208,and its protein was decreased to 0.372 ± 0.095 and 0.395 ± 0.077,significantly lower than in groups A and C (P < 0.05).In groups C and D,the level of bcl-xL mRNA was decreased to 0.514 ±0.183 and 0.509 ±0.168,and its protein was decreased to 0.447 ±0.097 and 0.410 ±0.089,significantly lower than than in groups A and B (P < 0.05).The growth of A549 cells in group D was inhabited strongly,and there was significant difference between group D with groups A,B and C ( P < 0.05 ).Conclusion HIF-1α siRNA combined with bcl-xL siRNA can significantly suppress the growth of 549 cells.
