Abstract
The article presents results of research on phages isolation of Klebsiella oxytoca bacterial species. Klebsiella oxytoca bacterial species are resistant organisms that cause many infectious diseases with severe course. In work there have been used bacterial strains obtained from the museum of the department MVEaVSE of Ulyanovsk SAA named after P.A. Stolypin. Main techniques were studied and tested for bacteriophages extraction, with additions for the studied microorganism. Extraction of Klebsiella oxytoca bacteriophage from bacterial cultures without exposure to inducing factors did not led to manifestation of free phage. Selection of bacteriophage by using inducing factors allowed to distinguish two phages active against Klebsiella oxytoca 1 and Klebsiella oxytoca 3. Selection of prophage from objects of environment allowed us to distinguish four strains of phages of Klebsiella oxytoca bacteria (indicator cultures Klebsiella oxytoca 1, Klebsiella oxytoca 2, Klebsiella oxytoca 10, Klebsiella oxytoca 24). One of the research stages was a study of basic biological properties that allow to note negative colonies distinctions of dedicated phages. Morphology of the phages, selected under the influence of UV-rays is characterized by greater turbidity and small size of negative colonies -0,51,0+0,5 mm. Phages obtained from environment objects are transparent, rounded, medium size 1,5-2,0+0,5 mm. Comparison of bacteriophages isolating techniques efficiency (expressed in the number of selected phages and girth breadth of the studied bacteria strains) allows us to conclude that each method is limited in use. All three methods are not universal for bacteriophages extraction of Klebsiella oxytoca bacterial species.
