Abstract

Theeffects ofbromobenzene on 6-aminolevulinate (ALA)dehydratase (EC4.2.1.24) wereexamined inrats. Theenzyme inthebonemarrow was irreversibly inhibited after 12hoursof treatment withbromobenzene (intraperitoneal ly) without anychange intheenzymeconcentration. Whenbromobenzene treatment was prolonged to72hours, theconcentrations oftheenzymeinthe bonemarrow andintheliver werereduced proportionally tothedecrease intheenzymeactivity. Neither theactivity northeconcentration ofALA dehydratase intheperipheral erythrocytes were reduced evenafter 72hours treatment withbromobenzene. Thesefindings indicate thatbromoben- zenedecreases ALA dehydratase activity ina biphasic manner;firstly, through an irreversible inhibition probably duetotheformation ofmercaptide withtheessential SHgroupsand,secondly, through areduced synthesis oftheenzyme.
