Abstract
OBJECTIVE: To establish a method of RP-HPLC for the determination of formonontin and calycasin from Radix Hedysari. METHODS: High-performance-liquid-chromatography (HPLC) on a Kromasil ODS column (5 microns, 4.6 mm x 250 mm) with water-methanol gradient elution, at column temperature of 25 degrees C, flow-rate of 1.0 ml/min and wavelength at 254 nm. RESULT: Linear ranges of formonontin and calycasin were 2.325 x 10(-3)-4.650 x 10(-2) micrograms and 2.350 x 10(-3)-4.700 x 10(-2) micrograms respectively. The average recoveries were 96.53% and 98.34% respectively, and relative standard deviations were 1.63% and 2.00%. CONCLUSION: The method was simple, reliable, accurate and practical.
