Abstract
Antibody-directed enzyme prodrug therapy (ADEPT) is a new approach to the treatment of solid cancers. In ADEPT, an antibody (or antibody fragment) is conjugated to an enzyme and is used to localise the enzyme at the site of the tumour. Once localised, the enzyme is used to activate a noncytotoxic prodrug to an active cytotoxic form. Therapy can proceed in either 2 or 3 phases. In the 2-phase system, the nonlocalised unbound antibody-enzyme conjugate is allowed to clear naturally from the body, and then the prodrug is administered. In the 3-phase system, clearance of unbound conjugate is accelerated by a ‘clearance’ antibody.
