Identification of RAPD markers linked to powdery mildew resistance gene Pm23 in wheat

Abstract

The Pm23 gene of wheat confers resistance to powdery mildew, which is one of the most serious diseases in many regions of the world. In this paper, bulked segregant analysis (BSA) was used to search for randomly amplified polymorphic DNA (RAPD) markers linked to Pm23 gene. 320 decamer primers were screened, and were one RAPD marker (OPE05(subscript 1100)) linked in coupling phase to Pm23. Using the F2 mapping population from a cross of Pm23×Chancellor, Pm23 was shown to be closely linked to the marker OPE05(subscript 1100) at a genetic distance of 10.65±3.25 cM. This marker can be conveniently used for marker-assisted selection in wheat breeding programs for the identification or pyramiding of Pm23 with other resistance genes.
