Abstract
In the experiment,tender leaf sheath of six sugarcane varieties was cultured in vitro to study the effects of different basic media,sugar concentrations,sugarcane varieties,sampling season and lighting culture conditions on the explant browning.The results indicated that the explant browning rate in N6 basic medium was the highest(35.44%)among three culture media;the explant browning degree was the largest when sucrose concentration was 40 g/L,while the average callus inducing rate was the highest with 30 g/L sucrose applied.As to different sugarcane varieties,the difference of explant browning rate among them reached highly significant level;the explant browning rate of Badila was the highest,while that of Funong 81-745 was the lowest.As compared to spring season,the average browning rate of six sugarcane varieties increased significantly while the average callus inducing rate decreased as sampling in winter season.Moreover,the average browning rate(35.44%)of explant in lighting culture(12 h/d)increased significantly as compared to dark culture.Therefore,sampling in spring season,adopting suitable basic culture medium and sugar concentration,and the tender leaf sheath of sugarcane was cultured in vitro under dark condition,could significantly decrease the browning rate of explant,and increase the allus inducing rate.
