Abstract

Objective To observe the brain distribution of bone marrow mesenchymal stem cells(MSCs),the effects on neurogenic functions and expression of apoptosis relative proteins Bcl-2 after intravenous transplanted into brain ischemic rats.Methods MSCs from SD rats were cultivated and proliferated in vitro then marked with CFSE.After that,MSCs were transplanted into middle cerebral artery occlusion(MCAO) models of SD rats by intravenous way.The brain distribution of MSCs and the expression of apoptosis relative proteins Bcl-2 were determined by immunohistochemistry and reverse transcription polymerase chain reaction(RT-PCR).Results The neurogenic functions of transplanted group significantly improved than those of control group on the day 6 after transplantation.The expressions of Bcl-2 protein at 6 h,12 h,48 h and 7 d time points increased significantly compared with those of control group,especially at 6 h and 48 h time points after transplantation.Relative contents of Bcl-2 mRNA of transplanted group at 48 h and 72 h also increased significantly compared with those of control group.Conclusion Bone marrow mesenchymal stem cells might exert the protective effects on brain ischemia and reperfusion injury by up-regulating Bcl-2.
