Apoptosis Induction of Bladder Cancer EJ Cells by Paclitaxel and Paclitaxel's Relationship to Gene Bcl-2

Abstract

Objective Investigate the apoptosis of bladder Cancer EJ cells and the mechanism induced by paclitaxel.Methods Bladder Cancer EJ cells cultured in vitro were treated by paclitaxel with different concentration for 12 h~72 h;Flow cytometry was used to examine the rate of apoptosis and the period of EJ cells;In situ hybridization was use to observe the variation of Gene Bcl-2 under Paclitaxel's influence.Results The period of EJ cells were stemed during the period of G2/M and the summit of apoptosis appeared when the concentration reached 100、1 000 nmol/L by flow cytometry.The rate of apoptosis grows with the growth of Paclitaxel's concentration.Gene Bcl-2 showed a clear deduction with the increase of Paclitaxel's concentration.Conclusion Paclitaxel can induce the apoptosis of bladder canner EJ cells,and the rate of apoptosis increase with the growth of the concentration of Paclitaxel;the change of Gene Bcl-2 was one of the reasons of cells apoptosis.
