Abstract
OBJECTIVE To study the effect of clarithromycin on the pharmacokinetics of theophylline in New Zealand rabbits.METHOD Healthy rabbits were given theophylline with either water or clarithromycin. Blood samples were collected at different time points.Plasma was assayed for thephylline by HPLC and various pharmacokinetic parameters were analyzed by 3P97 software.RESULTS As compared to controls,the t_ 1/2β,AUC and CL of theophylline were significantly changed in the rabbits with clarithromycin coadministration.The t_ 1/2β were increased to (4.90±1.16)h from (3.24±0.76)h(P0.01),the AUC were increased to (118.33±37.79)mg·h·L -1 from (75.02±22.41)mg·h·L -1(P0.01﹞,otherwise the CL were decreased to (0.13±0.041)·h -1 from (0.20±0.081)·h -1(P0.05).CONCLUSION The results indicate that clarithromycin may affect the t_ 1/2β, AUC and CL of theophylline.
