Abstract
Objective To express and purify the recombinant human interleukin 18 (rhIL 18) in prokaryotic system and to prepare the rabbit anti human interleukin 18 polyclonal antibody.Methods The cDNA of hIL 18 was amplified by RT PCR and was cloned into the expression vector pJW2 and transformed into the E.coli JM101. The purified recombinant protein was used to immune the rabbit for preparing polyclonal antibody.Results The cDNA of hIL 18 was sequenced and transformed into the E.coli. The recombinant human IL 18 (rhIL 18) was expressed efficiently in inclusion bodies and was successfully purified, the polyclonal antibody was successfully prepared with the purified recombinant human IL 18.Conclusion The rabbit anti human interleukin 18 polyclonal antibody can be used in further studies.
