Extracorporeal Anti-Hepatitis B Virus Effect of Oxymatrine with Different Administration Methods on 2.2.15 Cell Line and Its Mechanism

Abstract

Objective:To study the extracorporeal anti-hepatitis B virus effect of oxymatrine with different administration methods,intramus- cular injection,intravenous dripping and orally taken,and its mechanism.Methods:Taking HBsAg,HBeAg and survival rate of cells as indices,the anti-HBV effects of different administration methods of oxymatrine in 2.2.15 cell line in vitro were evaluated comprehen- sively and the copies of extracellular HBV DNA,intracellular HBV RNA and HBV DNA in supernatant were tested by fluorescent quantitative PCR.Results:For intramuscular injection of oxymatrine,the therapeutic index(T1)was more than 2,its ID_(50)on HBsAg and HBeAg was 2.5mg/ml and 3.2mg/ml respectively,CD_(50)was 16.6mg/ml,it showed obvious dose-dependent inhibition on intracellular and extracellular HBV DNA and intracellular HBV RNA(P0.01).Conclusion:Oxymatrine for intramuscular injection has obvious anti-HBV effect,the inhibitory position may be at the HBV DNA level.
