Abstract
Objective: At present, the detection of multidrug resistance depends on resected specimen but it can not be investigated in patients who were not operated on or got a long term chemotherapeutic treatment after operation. In this study, the relationship between p-glycoprotein expression in PBMC,CD 4+,CD8+ and tumor cells has been investigated. In order to obtain a new determination method for multidrug resistance, the changes of p-glycoprotein expression in PBMC,CD 4+,CD 8+ during chemotherapeutic stage has been analysed. Methods: The expression of p-glycoprotein in PBMC,CD 4+,CD8+ and tumor cells has been detected by means of immunohistochemestry assay in 23 cases, flow cytometry and immunohistochemestry assay in 28 cases to identify the relationship of p-glycoprotein between PBMC,CD 4+,CD8+ and tumor cells. P-glycoprotein in PBMC,CD 4+ and CD8+ was measured in 17 of 28 patients with chemotherapeutic treatment for six months. Results: ①The p-glycoprotein expression in PBMC,CD4+,CD8+ of patient with malignant tumor was higher than the control. ②A significance relationship of p-glycoprotein had been found between PBMC,CD 4+,CD8+ and tumor cells. ③ The p-glycoprotein expression in PBMC,CD 4+,CD 8+ increased significantly in chemotherapeutic group as compared with non-chemotherapeutic group. ④Flow cytometry assay was more sensitive than immunohistochemestry in detecting p-glycoprotein expression in PBMC,CD 4+,CD8+ . Conclusion:The detection of p-glycoprotein in PBMC,CD 4+,CD8+ provides a new method in the MDR determination. A relationship of p-glycoprotein has been found between PBMC,CD 4+,CD8+ and tumor cells.
