Abstract
Objective:To establish a HPLC assay method for the determination of Venlafaxine level in human serum.Methods:Using a mixture liquid of Acetonitrile:Methanol:Water:1mol/L-AmmoniumAcetate:5mol/L-NH4OH(50:350:100:5:1)as mobile phase,pH8.7,flow speed was lml/min,ODScolumn,the wave length of detection was229nm,Venlafaxine level was determined by means of external standard method.Results: A linearity was obtained from 25~700ngoml-l of Venlafaxine level in serum with a good correlation(r=:0.998).Detection limit was 2ngoml-l .The average extraction recovery was 96.59 % .The coefficients Of inter-day and intra-day variations were 3.44 %and3.77 % respectively.Conclusions: Using HPLC can detect Venlafaxine in serum well.
