Effect of Rosiglitazone on the ventricular remodeling and cardiac function of rabbit with acute myocardial infarction

Abstract

Objective To evaluate the effect of Rosiglitazone on rabbit ventricular remodeling and cardiac function after acute myocardial infarction(AMI) . Methods Thirty rabbits were randomly divided into:sham operation group(Sham) ,acute myocardial infarction(AMI) group,Rosiglitazone(Ros) group averagely. AMI model was established by ligation of the left anterior branches of the coronary artery,rabbits in Ros group were treated with Rosiglitazone 3mg/( kg·d ) by direct gastric gavage 6 hs after surgery,while AMI group and sham group were treated with equal amount of distille water. Left ventricular end-systolic dimension(LVESD) ,left ventricular end-diastolic dimension(LVEDD) ,ejection fraction(EF) ,shortening fraction(FS) were measured by heart ultrasound 4 weeks after surgery. Left ventricular weight(free wall+ interventricular septum) ,left ventricular weight index(left ventricle weight/volume quality) were examined with an electronic scale,level of tumor necrosis factor α(TNF-α) was determined by radioimmunoassay. Results The values of LVESD and LVEDD were higher,but the values of EF and FS were lower in AMI group than in sham group. The values of LVESD,LVEDD,Left ventricle weight,left ventricle weight index,and level of TNF-α in myocardium homogenate were significantly lower in Ros group than in AMI group,whereas EF was higher in Ros group than in AMI group. HE staining showed that the extent of myocardial cells fibration and inflammatory cells infiltration increased in AMI group,but decreased in Ros group. Conclusion Rosiglitazone can inhibit the ventricular remodeling and improve cardiac function after AMI,which may be related to the inhibition of the expression of inflammatory factors.
