Abstract

Objective To investigate the sensitization of the human lung adenocarcinoma cells A549 to cisplatin by Bcl-2 siRNA.Methods After transfection of bcl2-siRNA 48 hours according to the manufacturer's protocol,A549 cells were treated with different concentrations of cisplatin(0,0.5,1,2,3 μg/mL).48 hours later,the viability of cells was detected by cell counting Kit-8(CCK-8) assay according to the manufacturer's protocol.Real time RT-PCR was used to detect changes of Bcl-2 mRNA level after transfection of bcl2-siRNA.And western blot analysis was used to detect changes of Bcl-2 protein level after that.Results After transfection with bcl2-siRNA,the expression level of Bcl-2 mRNA in A549 cells decreased by 93.2%(P = 0.037),and the protein level decreased sharply,compared with the one transfected with negative control mimics.Meanwhile the sensitivity to cisplatin in A549 cells rose 69.8%(P = 0.024).Conclusion Bcl2-siRNA is successfully used to silence the expression level of Bcl-2 mRNA and protein,and to enhance the sensitivity to cisplatin in A549 cells line.
