Abstract
Objective To study the effects of acute methamphetamine(MA) exposure on lipopolysaccharide(LPS)-induced spleen cytokines IL-6,IL-10 and TNF-α in mice.Methods C57BL/6J mice were injected intraperitoneally with either MA(5 mg/kg) or normal saline(NS)(0.1 mL/kg);10 min following MA administration,the mice were given an immune stimulus of LPS(150 μg/kg) and their spleens were collected 30 min and 60 min after LPS treatment.Spleen samples were assessed for IL-6,IL-10 and TNF-α levels using ELISA kits.Results MA alone could increase the expressions of IL-6 and IL-10(P0.01).For IL-6 level,there was an MA+LPS interaction(P0.01) whereby MA increased the spleen level of IL-6 compared with LPS alone at 60 min post-LPS(P0.01).There was also an MA+LPS interaction for IL-10(P0.01) where MA increased the spleen level of IL-10 compared with LPS alone at 30 min post-LPS(P0.01).For TNF-α,there was no significant difference between MA treatment group and control group at 30 min and 60 min post-LPS(P=1.00).Comparison between the two different time points showed that the expressions of IL-6,IL-10 and TNF-α in the same group with LPS stimulation increased significantly at 60 min post-LPS(P0.01),whereas the levels of these cytokines without LPS stimulation did not differ significantly.Conclusion Acute MA exposure can significantly increase the expressions of LPS-induced spleenocytokines IL-6 and IL-10;however,it does not affect TNF-α expression.
